All the stats of a hero, like his knowledge and spellpower - what does each of them affect specifically? (as in which affects damage from DD spells, which affect mana...)
When you level up, will you get an increase in these stats in the same way each time, or is it random which ones go up?

Spell power affects, as its name implies, spell power: the amount of damage dealt by destructive magic, the effectiveness of blesses and curses, etc.
Knowledge affects your maximum mana, which is Knowledge * 10. For example, if you have 6 knowledge, you will have 60 mana.
I believe any hero has a certain chance to get any stat increased during level-up, but the exact probabilities are different for each class. Only one stat is increased by one with each level.

Knowledge also affects mana regeneration - it's [knowledge] mana per day (10% per day of your base mana, in other words), I believe, unless you have whatever ability doubles that. Knowledge also has the (indirect) effect of making the "summon creature" spell more useful since it costs 1 mana per creature summoned regardless of its tier.
You should download the manual on the site to look at the specific effects of spellpower on spells. Most direct damage spells have a damage calculation like 24 + (spellpower * 8) or something. Buffs usually last [spellpower] turns though in a few cases they last [spellpower/4] turns or something like that. A few summoning spells summon creatures based on spellpower (either more or stronger as power goes up).

Spellpower affects your damage and your bless/curse duration. For every point of spellpower, your curse/bless lasts that many turns on the target. Damage is calculated differently for each spell, which can be found in the manual. For example, Ice Bolt has a very high base damage, but its spellpower multiplier is low, so it will eventually be passed up by other spells, such as lightning bolt. Note that all spell effects (damage, blessing, and cursing) have their calculations and overall effects change as you progress from having no skill with the school to expert level.

Overall, knowledge is for longevity, and in previous games was often considered a hindrence to casters when brought up in excess. However, with the addition of the summon creatures spell and the innate mana regeneration addition, knowledge has gone up considerably in value.
